“Silencing Mesothelin: A Novel Approach to Treatment of Mesothelioma”

Mesothelin, a plasma membrane antigen expressed in significantly high levels in mesothelioma patients has been studied as marker for diagnosis of this malignancy as well as a target for monoclonal antibodies. We, however, have tried to target this protein from a different perspective: we have produced promising preliminary data showing that inhibition of the expression of mesothelin with the novel technology of short-interfering RNA (siRNA) causes a significant reduction in the proliferation rate of mesothelioma cells while decreasing their invasiveness. Furthermore, we are now progressing towards developing a “replication incompetent lentivirus” capable of expressing anti-mesothelin siRNA and testing its efficiency/specificity in causing cytotoxicity against mesothelioma cells in-vitro and tumor regression in-vivo. If promising results are obtained in future steps, this project can lead to development of a novel gene therapy agent with enhanced specificity for targeting mesothelioma….
